A rapid and sensitive separation of retinol and retinyl palmitate using a small, disposable bonded-phase column: kinetic applications.

چکیده
A method utilizing small disposable C18 bonded-phase columns has been developed to separate retinol and retinyl palmitate mixtures into their individual components in high yield and purity. Up to ten mixtures can be processed in 1 hr and the columns are reusable after suitable washing. Although the method was developed with standard retinoid mixtures, it was shown that it is also applicable to the assay of the kinetics of both a bile salt-stimulated human milk lipase-catalyzed hydrolysis reaction and acyl transfer reaction. This rapid, accurate, and inexpensive method is complementary to other chromatographic techniques, especially in kinetic investigations, and enables one to detect these fluorescent retinoids in quantities as small as 2 picomoles. --O'Connor, C. Determination of retinyl esters and retinol in serum or plasma by normal-phase liquid chromatography: method and applications.
Retinol and retinyl esters are measured in serum or plasma samples by gradient, normal-phase, adsorption "high-performance" liquid chromatography, with ultraviolet detection at 325 nm. The four major circulating retinyl esters in humans (esters of palmitate, stearate, oleate, and linoleate) are coeluted as a single peak. Retinyl acetate is included as an internal standard, to correct for variab...
متن کاملRetinol esterification by microsomes from the mucosa of human small intestine. Evidence for acyl-Coenzyme A retinol acyltransferase activity.
The mechanism of the intestinal esterification of retinol has been obscure. Recently, an acyl-Coenzyme A (CoA):retinol acyltransferase (ARAT) was found in rat intestinal microsomes, and experiments were therefore conducted to determine whether a corresponding enzyme exists in human small intestine. When microsomes were incubated with [3H]retinol and palmitoyl-CoA, or retinol and [1-14C]palmitoy...
متن کاملSimultaneous determination of fat-soluble vitamins and provitamins in dairy products by liquid chromatography with a narrow-bore column.
A reversed-phase high-performance liquid chromatographic method is described for the simultaneous determination of vitamins A, D2, D3, E and K1, retinyl acetate, retinyl palmitate, tocopherol acetate, ergosterol and 7-dehydrocholesterol in milk and butter. Narrow-bore columns are recommended because this alternative provides a good separation and efficiency, plus greater economy and sensitivity...
متن کاملAn LC/MS/MS method for stable isotope dilution studies of β-carotene bioavailability, bioconversion, and vitamin A status in humans.
Isotope dilution is currently the most accurate technique in humans to determine vitamin A status and bioavailability/bioconversion of provitamin A carotenoids such as β-carotene. However, limits of MS detection, coupled with extensive isolation procedures, have hindered investigations of physiologically-relevant doses of stable isotopes in large intervention trials. Here, a sensitive liquid ch...
متن کاملRetinyl ester hydrolysis and retinol efflux from BFC-1beta adipocytes.
Adipose tissue is an important storage depot for retinol, but there are no data regarding retinol mobilization from adipose stores. To address this, dibutyryl cAMP was provided to murine BFC-1beta adipocytes and its effects on retinol efflux assessed. High performance liquid chromatography analysis of retinol and retinyl esters in adipocytes and media indicated that cAMP stimulated, in a time- ...
